Recombinant grass pollen allergens effectively reduced allergic rhinitis symptoms and medication needs. This immunotherapy approach also improved quality of life and induced beneficial immune responses, showing promise for treating hay fever.
Area of Science:
- Immunology
- Allergology
- Biotechnology
Background:
- Allergen-specific immunotherapy traditionally uses natural extracts to manage allergic responses.
- Recombinant DNA technology allows for the production of specific allergens, enhancing research into improved immunotherapy.
Purpose of the Study:
- To evaluate the efficacy of a 5-recombinant grass pollen allergen mixture in alleviating symptoms and reducing medication requirements in grass pollen-allergic individuals.
- To assess the impact on quality of life and immune responses.
Main Methods:
- A randomized, double-blind, placebo-controlled subcutaneous immunotherapy trial was conducted.
- Participants had allergic rhinoconjunctivitis, with or without asthma.
- Key measures included a symptom-medication score, quality of life questionnaire, conjunctival provocation tests, and specific antibody analysis.
Main Results:
- Significant reductions in both symptoms and medication use were observed in the recombinant allergen group compared to placebo.
- Clinically relevant improvements in quality of life were reported.
- Strong allergen-specific IgG(1) and IgG(4) antibody responses were induced, even in some individuals not initially sensitized to specific components like Phl p 5.
Conclusions:
- A vaccine composed of recombinant allergens is a safe and effective treatment for allergic rhinitis symptoms.
- Clinical benefits correlate with immune response modulation, including increased IgG(4) and decreased IgE, suggesting the induction of regulatory T cells.

Antibody Structure
Antibodies, also known as immunoglobulins (Ig), are essential players of the adaptive immune system. These antigen-binding proteins are produced by B cells and make up 20 percent of the total blood plasma by weight. In mammals, antibodies fall into five different classes, which each elicits a different biological response upon antigen binding.

Antiasthma Drugs: Mast Cell Stabilizers and Anti-IgE Drugs
Mast cell stabilizers, such as cromolyn (also known as sodium cromoglycate) and nedocromil (Tilade), are effective drugs in asthma management. These stabilizers hinder histamine release by skillfully obstructing the activation of mast cells and other cellular entities. Notably, they navigate this task without...
