Osmoregulation in Fishes02:32

Osmoregulation in Fishes

53.0K
When cells are placed in a hypotonic (low-salt) fluid, they can swell and burst. Meanwhile, cells in a hypertonic solution—with a higher salt concentration—can shrivel and die. How do fish cells avoid these gruesome fates in hypotonic freshwater or hypertonic seawater environments?

Cell Lines01:16

Cell Lines

10.1K
A cell line is a population of cells grown in vitro that can be subcultured over several generations. Normal cells cease to divide after a certain number of cell divisions, a process known as replicative senescence. This number, called the Hayflick limit, was conceptualized by Leonard Hayflick in 1961 when he observed that fetal cells grown in culture could only divide 40-60 times. This limit is due to the shortening of the telomeres during each round of cell division, preventing cell division...

Proteomics

9.4K
A proteome is the entire set of proteins that a cell type produces. We can study proteomes using the knowledge of genomes because genes code for mRNAs, and the mRNAs encode proteins. Although mRNA analysis is a step in the right direction, not all mRNAs are translated into proteins.
Proteomics is the study of proteomes' function. It involves the large-scale systematic study of the proteome to denote the protein complement expressed by a genome. Scientist Mark Wilkins coined the term...

Lossless Lines01:23

Lossless Lines

566
In electrical engineering, a lossless transmission line is characterized by a purely imaginary propagation constant and a resistive characteristic impedance. The ABCD parameters, which describe the relationship between the input and output voltages and currents, indicate an equivalent π circuit with an imaginary series impedance and a shunt admittance. This results in a transmission line that, when the product of the phase constant (beta) and the length of the line is less than pi, exhibits...

Lossy Lines and Overvoltages01:22

Lossy Lines and Overvoltages

357
Transmission-line series resistance and shunt conductance cause three primary effects: attenuation, distortion, and power losses.
Attenuation
When constant series resistance and shunt conductance are present, voltage and current equations are modified. The propagation constant indicates that voltage and current waves consist of both forward and backward traveling components. These waves attenuate as they propagate, with the attenuation factor related to the resistance and conductance. In a...

Electric Field Lines01:25

Electric Field Lines

9.4K
The three-dimensional representation of the electric field of a positive point charge requires tracing the electric field vectors, whose lengths decrease as the square of their distance from the charge and which point away from the charge at each point. This vector field is no doubt challenging to visualize. The visualization of electric fields becomes quickly intractable as the number of charges increases.
The solution to this problem is to use electric field lines, which are not vectors but...

Summary

Misidentification of fish cell lines is a concern. A new proteomic approach using protein expression signatures (PES) offers a reliable method for routine fish cell line identification and contamination screening.

Area of Science:

  • Aquaculture
  • Cell Biology
  • Proteomics

Background:

  • Fish cell line cultures are prone to cross-contamination due to simple growth requirements.
  • Misidentification and cross-contamination of fish cell lines can compromise scientific results and reproducibility.
  • Existing methods for intra-species fish cell line identification are often complex and time-consuming.

Purpose of the Study:

  • To develop a simple and reliable proteomic method for identifying fish cell lines.
  • To establish protein expression signatures (PES) for distinguishing between different fish cell lines.
  • To provide a routine screening tool for fish cell line authentication.

Main Methods:

  • Utilized two-dimensional gel electrophoresis (2-DE) to analyze protein expression profiles.

  • Developed protein expression signatures (PES) for various fish cell lines.
  • Created an electronic databank to store and compare PES profiles.
  • Main Results:

    • Protein expression signatures (PES) successfully differentiated between fish cell lines.
    • Higher concordance was observed among cell lines from the same fish species (e.g., rainbow trout).
    • Similar concordance was found in cell lines derived from the same tissues within a species.

    Conclusions:

    • A proteomic approach using PES is a valuable and reliable technique for fish cell line identification.
    • This method can aid in discerning the origins of cell lines and detecting intra-species contamination.
    • The developed PES profiles and databank can support routine cell line authentication in research.
