DNAzymes targeting isocitrate lyase (ICL) mRNA effectively silenced the gene in Mycobacterium tuberculosis. Combining DNAzymes with isoniazid (INH) reduced latent tuberculosis infection and bacterial survival in macrophages.

Area of Science:

  • Molecular Biology
  • Microbiology
  • Biochemistry

Background:

  • Latent Mycobacterium tuberculosis infection poses a significant challenge to tuberculosis treatment.
  • Isocitrate lyase (ICL) is crucial for M. tuberculosis metabolism during the latent phase.

Purpose of the Study:

  • To investigate the efficacy of sequence-specific DNAzymes targeting ICL mRNA.
  • To evaluate the impact of DNAzyme treatment on ICL expression and M. tuberculosis survival.

Main Methods:

  • Designed and tested five DNAzymes for specific cleavage of icl mRNA in vitro.
  • Treated virulent M. tuberculosis with DNAzyme DZ4 and subinhibitory isoniazid (INH).
  • Assessed ICL expression and M. tuberculosis survival in macrophages.

Related Experiment Videos

Main Results:

  • Four of five DNAzymes (DZ1, DZ3, DZ4, DZ5) demonstrated efficient and specific icl mRNA cleavage.
  • Treatment with DZ4 and INH significantly reduced ICL expression and M. tuberculosis survival in macrophages.
  • Combined treatment showed no significant impact on M. tuberculosis growth in vitro.

Conclusions:

  • DNAzyme-mediated silencing of the icl gene is a promising strategy against latent tuberculosis.
  • Using INH enhances DNAzyme uptake by M. tuberculosis, improving therapeutic potential.
