Summary

Medulloblastoma cell proliferation is high, but survival depends more on radiation therapy sensitivity than growth rate. This study correlated cell kinetics with clinical behavior in medulloblastoma patients.

Background:

  • Correlating cell kinetics with clinical behavior in medulloblastoma.
  • Analyzing 22 primary and 10 recurrent/metastatic medulloblastomas from 30 patients.

Observation:

  • Bromodeoxyuridine (BUdR) and tritiated thymidine were used to determine the labeling index (LI).
  • Mean LI of 11.7% indicated high proliferative potential in medulloblastomas.
  • Higher LI observed in tumors from younger patients and in the cerebellar hemisphere.

Findings:

  • LI did not correlate with patient survival time.
  • S-phase duration ranged from 6.1-11.3 hours (mean 8.0 hours).
  • Potential doubling time was 25-82 hours; actual doubling time was 20-24 days, with an estimated cell loss factor of 0.90.

Implications:

  • High LI suggests rapid medulloblastoma growth.
  • Patient survival appears more influenced by radiation therapy response than tumor proliferation rate.
  • Further research into medulloblastoma growth dynamics and treatment sensitivity is warranted.
