Summary

Molecular margin analysis using CDKN2A methylation detected preneoplastic lesions in hepatocellular carcinoma (HCC) patients. This method shows promise for improving prognosis in HCC patients after surgery.

Area of Science:

  • Oncology
  • Molecular Biology
  • Surgical Pathology

Background:

  • Positive surgical margins in hepatocellular carcinoma (HCC) correlate with postsurgical recurrence.
  • Molecular margin analysis offers higher sensitivity for detecting preneoplastic lesions than traditional histological examination.

Purpose of the Study:

  • To assess the feasibility and clinical utility of methylation-based molecular margin analysis in HCC.
  • To investigate the CDKN2A methylation status in surgical margins of HCC patients.

Main Methods:

  • A nested bisulfite-specific methylation-polymerase chain reaction (BS-MSP) protocol was employed.
  • CDKN2A methylation status was analyzed in surgical margins, tumor tissues, and adjacent nonmalignant tissues from 20 HCC patients.

Related Experiment Videos

Main Results:

  • CDKN2A methylation was detected in 35% (7 of 20) of histologically negative margins.
  • Methylation patterns in resection margins may aid in understanding the cellular origins of recurrent carcinoma.

Conclusions:

  • Methylation-based molecular surgical margin analysis is a feasible and promising tool for HCC prognosis.
  • This approach can enhance the detection of residual disease and improve patient outcomes after hepatectomy.
