Summary

This study demonstrates that insulin triggers the release of kinins, which are signaling molecules. These kinins cause uterine contractions, providing direct evidence for insulin

Area of Science:

  • Pharmacology
  • Endocrinology
  • Physiology

Background:

  • Kinins are vasoactive peptides implicated in various physiological processes.
  • The role of kinins in insulin-mediated physiological responses remains incompletely understood.

Purpose of the Study:

  • To investigate whether hyperglycemia or insulin administration in vivo leads to kinin release.
  • To characterize the uterine contractile responses to insulin and glucose in rats.

Main Methods:

  • Utilized the rat uterus in situ as a bioassay for kinins.
  • Administered bradykinin, glucose, and insulin intravenously.
  • Employed bradykinin receptor antagonists (HOE 140), angiotensin-converting enzyme (ACE) inhibitors (captopril), oxytocin antagonists (atosiban), and plasma kallikrein inhibitors (SBTI).

Main Results:

  • Bradykinin induced uterine contractions, blocked by the bradykinin B2 receptor antagonist HOE 140.
  • Intravenous glucose infusion led to hyperglycemia and increased uterine contractility, abolished by HOE 140.
  • Insulin administration, with or without glucose, induced uterine contractions suppressed by HOE 140 or SBTI.

Conclusions:

  • These findings provide direct evidence that insulin administration induces the release of kinins.
  • The study implicates kinin B2 receptors and plasma kallikrein in insulin-mediated uterine responses.
