Summary

Transforming growth factor beta (TGF-beta) activation occurs early in cochlear immune responses, decreasing as the response resolves. This suggests early intervention may prevent cochlear fibrosis.

Background:

  • Transforming growth factor beta (TGF-beta) is a key mediator of fibrogenesis.
  • The role of TGF-beta in cochlear immune responses and fibrosis is not well understood.

Purpose of the Study:

  • To investigate the involvement of TGF-beta during cochlear immune responses.
  • To determine the temporal expression of TGF-beta and latency-associated peptide (LAP) in the cochlea during an adaptive immune response.

Main Methods:

  • An adaptive immune response to antigen was induced in mice.
  • Cochleas were analyzed using immunocytochemistry for TGF-beta and LAP at various time points (3-48 hours and 7 days post-initiation).

Main Results:

  • LAP was detected in normal cochleas, while TGF-beta was found in inflammatory cells early (3 hours) after antigen challenge.

  • A rapid decrease in LAP and increase in TGF-beta expression indicated TGF-beta activation during the immune response.
  • These changes persisted for 48 hours before returning to normal by 7 days.
