Transforming growth factor beta regulates cyclooxygenase-2 in glomerular mesangial cells

Transforming growth factor beta (TGFbeta) upregulates cyclooxygenase-2 (COX-2) and prostaglandin E synthase (mPGES-1) in rat kidney cells. This TGFbeta-induced COX-2 expression affects prostaglandin production but not collagen synthesis.

- COX-2 and mPGES-1 expression were analyzed using Northern blot and real-time PCR.
- Luciferase reporter assays and gel shift analyses were employed to study gene transcription and transcription factor activation (AP-1, NF-kappaB).
- Prostaglandin E2 (PGE2) and thromboxane B2 (TxB2) production were measured by enzyme immunoassay.
Main Results:
- TGFbeta1 treatment significantly induced COX-2 mRNA and mPGES-1 expression in rat mesangial cells.
- TGFbeta1 increased PGE2 production threefold, an effect reversed by COX-2 inhibition.
- TGFbeta1 enhanced COX-2 gene transcription by approximately 50% and increased COX-2 mRNA stability.
- Activation of transcription factors AP-1 and NF-kappaB was observed following TGFbeta1 treatment.
- Neither collagen type I mRNA levels nor net collagen production were affected by co-treatment with NS398.
Conclusions:
- TGFbeta stimulates the expression of COX-2 and mPGES-1 in rat mesangial cells.
- TGFbeta regulates COX-2 at both the transcriptional and post-transcriptional (mRNA stability) levels.
- While TGFbeta-induced COX-2 influences prostaglandin production, it does not appear to mediate downstream collagen synthesis in this model.
