A novel DNA-binding factor, ECIF-1, recognizes a specific octamer motif in the human beta-interferon promoter. Its binding decreases with cell differentiation, impacting interferon inducibility and gene repression.

Area of Science:

  • Molecular Biology
  • Gene Regulation
  • Cell Differentiation

Background:

  • ECIF-1 (embryonal carcinoma-specific interferon factor 1) is a DNA-binding factor identified in murine embryonal carcinoma cells.
  • This factor recognizes a specific region within the human beta-interferon promoter.

Purpose of the Study:

  • To characterize the DNA-binding properties of ECIF-1.
  • To investigate the role of ECIF-1 in the regulation of beta-interferon gene expression during cell differentiation.

Main Methods:

  • Site-directed mutagenesis to identify critical nucleotides within the ECIF-1 binding site.
  • Electrophoretic mobility shift assays (EMSAs) to assess protein-DNA interactions.
  • In vitro transcription assays using deleted beta-interferon promoter elements.

Related Experiment Videos

  • Nuclear extracts from undifferentiated and differentiated P19 cells were utilized.
  • Main Results:

    • The promoter region between -112 and -93 of the human beta-interferon gene is sufficient for ECIF-1 binding.
    • The critical binding motif for ECIF-1 was identified as the octamer ATTTACAT.
    • ECIF-1 binding activity diminishes upon P19 cell differentiation, coinciding with increased interferon inducibility.
    • Undifferentiated P19 nuclear extracts exhibit a repressing activity dependent on the ECIF-1 site, which is absent in differentiated cells.

    Conclusions:

    • ECIF-1 is a distinct DNA-binding factor, not interferon-regulatory factor 1 or 2, that binds to an octamer motif in the beta-interferon promoter.
    • ECIF-1 plays a role in repressing beta-interferon gene expression in undifferentiated cells.
    • The regulation of ECIF-1 binding and its associated repressing activity is linked to cell differentiation and the onset of interferon inducibility.
