HIV-infected cells utilize reverse transcription complexes (RTCs) for viral DNA synthesis. Defective Vif protein in HIV leads to abnormal RTCs with increased mutations in reverse transcription products within newly infected cells.

Background:

  • Reverse transcription (RTn) in HIV occurs within a nucleoprotein complex called the reverse transcription complex (RTC).
  • RTCs contain reverse transcriptase (RT) and integrase (IN) and exhibit heterogeneity in size and density.
  • The Vif protein plays a role in HIV replication, particularly in non-permissive cells.

Purpose of the Study:

  • To investigate the characteristics of RTCs in HIV infections, focusing on the impact of Vif deficiency.
  • To analyze the sedimentation properties and mutation levels of RTCs produced by Vif-deficient (Deltavif) HIV.
