Summary
We developed a novel fluorescent dye, 2,5-Bis[1-(4-N-methylpyridinium)ethen-2-yl)]-N-methylpyrrole ditriflate (PEPEP), for mitochondrial imaging. This nontoxic probe is highly effective for confocal fluorescence microscopy in various cell types.
Area of Science:
- Cell Biology
- Biochemistry
- Microscopy
Background:
- Mitochondrial probes are essential tools for cellular research.
- Existing probes may have limitations such as toxicity or potentiometric dependence.
- Novel fluorescent dyes are needed for advanced imaging techniques.
Purpose of the Study:
- To introduce and characterize a new fluorescent dye, 2,5-Bis[1-(4-N-methylpyridinium)ethen-2-yl)]-N-methylpyrrole ditriflate (PEPEP), as a mitochondrial probe.
- To evaluate the efficacy and safety of PEPEP for confocal fluorescence microscopy.
Main Methods:
- Synthesis and characterization of the PEPEP chromophore.
- Cytotoxicity assays in mammalian and yeast cell cultures.
- Intracellular localization studies using confocal fluorescence microscopy.
Main Results:
- PEPEP demonstrated minimal cytotoxicity in tested cell lines.
- The dye effectively localized to mitochondria in both mammalian and yeast cells.
- PEPEP exhibited strong fluorescence emission suitable for confocal microscopy.
Conclusions:
- PEPEP is a novel, nontoxic, and nonpotentiometric mitochondrial probe.
- It serves as an efficient and valuable alternative to current mitochondrial stains for confocal fluorescence microscopy.
