Cork suberin, a plant polymer, was depolymerized to reveal glycerol-derived lipids. These findings suggest suberin is a polymeric glycerolipid with roles similar to biological membrane lipids.
Area of Science:
- Plant biochemistry
- Polymer chemistry
- Lipidomics
Background:
- Cork suberin is a complex plant biopolymer.
- Its exact polymeric structure and monomeric composition remain areas of active research.
- Understanding suberin's structure is crucial for its potential applications.
Purpose of the Study:
- To partially depolymerize cork suberin.
- To identify the monomeric and oligomeric components released.
- To elucidate the polymeric nature of suberin.
Main Methods:
- Partial depolymerization of cork suberin using methanolysis.
- Methanolysis catalyzed by calcium hydroxide.
- Analysis of the methanolisate using electrospray ionization tandem mass spectrometry (ESI-MS/MS) with lithium adducts.
Main Results:
- The reaction solubilized a mixture of monomers and oligomers.
- Glycerol-derived dimeric and trimeric esters were identified.
- Specific glycerol esters identified include monoacylglycerols, diglycerol diesters, and diacylglycerols, incorporating various diacids and hydroxyacids.
- The identified alpha,omega-diacids and omega-hydroxyacids correspond to known suberin monomers.
Conclusions:
- Suberin is a polymeric lipid derived from glycerol.
- The identified components confirm suberin's complex esterified structure.
- Suberin shares characteristics with glycerolipids found in biological membranes, suggesting related functional roles.
