Summary

Novobiocin induces Bacillus subtilis recF gene expression, but this effect is dependent on the gyrB gene and independent of the SOS response. The RecF protein does not regulate its own expression.

Area of Science:

  • Microbiology
  • Molecular Biology
  • Genetics

Background:

  • The recF gene in Bacillus subtilis is located between the dnaN and gyrB genes.
  • A specific mutation, recF15, alters a glutamate residue to lysine in the RecF protein.

Purpose of the Study:

  • To investigate the in vivo regulation of the Bacillus subtilis recF gene.
  • To determine the effect of novobiocin on recF gene expression.
  • To ascertain the relationship between recF regulation, the gyrB gene, and the SOS response.

Main Methods:

  • Utilized a transcriptional fusion of the recF gene to the xylE reporter gene.
  • Quantified mRNA production to assess recF gene expression levels.
  • Compared recF expression in wild-type and gyrB mutant strains of Bacillus subtilis.

Main Results:

  • Novobiocin treatment resulted in a four-fold increase in recF gene expression.
  • This induction by novobiocin was not observed in a gyrB mutant strain.
  • The enhancement of recF expression by novobiocin was found to be independent of the SOS response.

Conclusions:

  • recF gene expression in Bacillus subtilis is regulated in response to novobiocin.
  • This regulation is dependent on the gyrB gene.
  • The RecF protein itself does not appear to be involved in the autoregulation of recF expression.
