Background:

  • Candida albicans is a yeast that has been implicated as a potential dental caries pathogen.
  • Evidence supporting the specific role of Candida albicans in caries progression remains limited.
  • Significant yeast presence within a lesion is considered a prerequisite for its role in caries pathology.

Purpose of the Study:

  • To investigate the presence and extent of Candida albicans in human dentinal caries lesions.
  • To evaluate the suitability of different staining methods for detecting yeasts in carious dentin.

Main Methods:

  • Human carious teeth (n=10) were sectioned and stained using Gram, Giemsa, Periodic Acid-Schiff (PAS), and immunohistochemistry with a Candida albicans-specific antibody.
  • A total of 120 sections were analyzed across the different staining techniques.

Main Results:

  • Extensive bacterial invasion and positive PAS staining were observed in all samples.
  • Immunohistochemistry detected only a few weakly positive sections (3.3%) with minimal Candida albicans cells on the lesion surface.
  • Positive identification of Candida albicans based on cell morphology was not achievable.

Conclusions:

  • The findings do not support the hypothesis that Candida albicans plays a significant role in dentinal caries pathology.
  • Periodic Acid-Schiff (PAS) staining demonstrated an unspecific nature and was deemed unsuitable for detecting yeasts in carious tooth samples.
