Summary

Human endogenous retroelements express functional reverse transcriptase (RT) proteins in the skin. Psoriasis is associated with increased RT expression and activity, suggesting a role in skin pathology.

  • Specific assays optimized for different retroviral types (C-type and D-type).
  • Main Results:

    • RT proteins were detected in normal skin epidermis and dermis, with a prominent 57 kDa band.
    • Psoriatic lesions showed increased RT-positive cells in both epidermal and dermal compartments, notably in Munro's abscesses.
    • RT activity was 2-3 times higher in psoriatic skin extracts compared to normal skin.

    Conclusions:

    • Functional RT proteins are expressed by active endogenous retroelements in the skin.
    • While basal RT expression is present in normal skin, activity is low.
    • Psoriasis is characterized by elevated RT-positive cells and significantly increased RT activity, potentially linked to keratinocyte hyperproliferation and inflammation.
    • Endogenous retroelements may have a significant role in skin physiology and pathology, warranting further investigation.
