We developed TomoJ, a new ImageJ plugin for transmission electron tomography (3D electron microscopy). This freeware enhances 3D structural analysis of subcellular components, bridging microscopy techniques.
Area of Science:
- Cell Biology
- Microscopy
- Structural Biology
Background:
- Transmission electron tomography (TET) is a powerful 3D imaging technique in electron microscopy.
- TET bridges the resolution gap between high-resolution methods and optical microscopy.
- Subcellular structure analysis benefits from TET's detailed insights.
Purpose of the Study:
- Introduce TomoJ, a novel software tool for transmission electron tomography.
- Provide a user-friendly ImageJ plugin for enhanced 3D electron microscopy data processing.
- Facilitate advanced analysis of subcellular structures using TET.
Main Methods:
- Developed TomoJ as a Java plugin for ImageJ, a standard image processing platform.
- Integrated advanced reconstruction algorithms: Algebraic Reconstruction Technique (ART) and Simultaneous Iterative Reconstruction Technique (SIRT).
- Included Weighted Back-Projection (WBP) for versatile volume reconstruction.
Main Results:
- TomoJ offers a streamlined interface for aligning, reconstructing, and combining tomographic volumes.
- The software supports state-of-the-art and commonly used reconstruction algorithms.
- Facilitates comprehensive analysis of 3D electron microscopy datasets.
Conclusions:
- TomoJ is specifically engineered for electron tomography applications.
- The software is freely available as a Java plugin for ImageJ.
- TomoJ empowers researchers with advanced, accessible tools for subcellular structural analysis.

The technique was invented in the 1970s and is based on the principle that as X-rays pass through the body, they are absorbed or reflected at different levels. In the technique, a patient lies on a motorized platform while a computerized axial tomography (CAT) scanner rotates...
