Overexpression of E2F1 and cyclin E (CCNE) is linked to clear cell renal cell carcinoma (ccRCC) progression. Cyclin E levels increase in tumors and surrounding tissues, correlating with poorer differentiation.
Area of Science:
- Oncology
- Molecular Biology
- Biochemistry
Background:
- Clear cell renal cell carcinoma (ccRCC) is the most common subtype of kidney cancer.
- The cell cycle regulators E2F1 and cyclin E (CCNE) play critical roles in cell proliferation and are often dysregulated in cancer.
Purpose of the Study:
- To investigate the relationship between E2F1 and CCNE expression in ccRCC tissues.
- To explore the potential role of CCNE in ccRCC tumorigenesis and its correlation with tumor differentiation.
Main Methods:
- Quantitative analysis of E2F1 and CCNE mRNA and protein levels in ccRCC, peritumoral, and cancer-free kidney tissues.
- Assessment of correlations between E2F1, CCNE, and tumor differentiation status.
Main Results:
- E2F1 overexpression was associated with increased CCNE mRNA and protein levels in ccRCC tissues compared to controls.
- Elevated total cyclin E was observed in peritumoral tissues, suggesting paracrine signaling.
- Positive correlations between E2F1 and CCNE were found in normal and peritumoral tissues but not in ccRCCs.
- CCNE overexpression correlated with decreased tumor differentiation.
Conclusions:
- E2F1 and CCNE are significantly altered in ccRCC, with CCNE overexpression linked to poorer tumor differentiation.
- The findings suggest a role for CCNE in ccRCC progression and highlight potential cross-talk between tumor and surrounding tissues.

The Retinoblastoma Gene
The first-ever tumor suppressor gene called Rb was identified in retinoblastoma - a rare eye tumor in children. In inherited forms of the disease, a child inherits one defective copy of the Rb gene, which predisposes them to retinoblastoma. However,...

M-Cdk Drives Transition Into Mitosis
Cyclin-dependent kinases, or Cdks, work in concert with cyclins to control cell cycle transitions. M-Cdk, a complex of Cdk1 bound to M cyclin, is a well-known example of this coordinated control that drives the transition from the G2 to the M phase.
M cyclin...
