Selective cyclooxygenase-2 inhibitors like Celecoxib and Parecoxib can manage orthodontic pain without hindering tooth movement. Rofecoxib, however, significantly slowed dental movement in rats.

- To compare the effects of different COX-2 inhibitors (Rofecoxib, Celecoxib, Parecoxib) on orthodontic tooth movement in a rat model.
- To determine if these selective COX-2 inhibitors interfere with the biological processes of tooth movement.
Main Methods:
- Twenty-eight Wistar rats were divided into four groups: Rofecoxib, Celecoxib, Parecoxib, and saline control.
- Orthodontic tooth movement was induced using coil springs.
- Selective COX-2 inhibitors or saline were administered via gingival injections over 5 days.
- Tooth movement was quantified using lateral cranial teleradiographs after 10 days.
Main Results:
- Control rats exhibited mesial tooth displacement of 0.33 ± 0.07 mm.
- Rofecoxib treatment resulted in no measurable tooth movement.
- Celecoxib and Parecoxib groups showed tooth movement of 0.42 ± 0.09 mm and 0.22 ± 0.04 mm, respectively.
- Statistical analysis revealed significant differences between groups (P < 0.004).
Conclusions:
- Celecoxib and Parecoxib appear to be suitable for managing orthodontic pain without compromising tooth movement.
- Rofecoxib demonstrated an inhibitory effect on orthodontic tooth movement.
- Selective COX-2 inhibitors have differential effects on orthodontic treatment efficacy.
