    Background:

    • Junin virus is a significant human pathogen belonging to the arenavirus family.
    • Understanding the replication and RNA composition of Junin virus is crucial for developing antiviral strategies.

    Purpose of the Study:

    • To characterize the RNA species of Junin virus.
    • To investigate the effect of actinomycin D on Junin virus RNA labeling.

    Main Methods:

    • Junin virus was propagated in BHK-21 cells and labeled with [3H]uridine.
    • High purification of the virus was achieved through differential and isopycnic centrifugation.
    • RNA extraction and analysis were performed using phenol and polyacrylamide gel electrophoresis.

    • Junin virus RNA comprises four large species (33, 28, 18S) and three small species (4, 5, 5.5S).
