Summary

A novel Human Leukocyte Antigen B allele, identified in a Chinese Han individual, was characterized through sequencing. This new HLA-B allele was confirmed as an extension of B*2736.

Immunoglobulin Gene Sequence Analysis In Chronic Lymphocytic Leukemia: From Patient Material To Sequence Interpretation

  • Allele-specific primers were used to amplify exons 2-5, including introns 2-4, of the novel HLA-B*27 allele.
  • Bidirectional sequencing was performed to identify nucleotide differences compared to HLA-B*270401 and other related alleles.
  • Main Results:

    • A novel HLA-B allele, distinct from HLA-B*270401, was identified with 10 nucleotide changes in exons 3-4 and specific mutations/deletions in introns 2-4.
    • Intron 2 showed homology with HLA-B*2706, while introns 3 and 4 shared sequences with HLA-B*070201.
    • The novel allele sequence (1,815 bp) was submitted to GenBank (accession number DQ915176).

    Conclusions:

    • The novel HLA-B allele represents a new variant within the HLA-B*27 family.
    • The genetic characterization provides valuable data for the HLA database and immunogenetic research.
    • The allele was officially confirmed as an extension of B*2736 by the WHO Nomenclature Committee.
