Summary

This study found specific Human Leukocyte Antigen (HLA) genes are associated with pemphigus susceptibility and protection in Morocco. These findings confirm a genetic predisposition to pemphigus in the Moroccan population.

Background:

  • Pemphigus is an autoimmune blistering disease targeting keratinocyte adhesion molecules.
  • Human Leukocyte Antigen (HLA) class II genes, especially DR4 and DR14, show known associations with pemphigus globally.
  • Genetic predisposition to pemphigus has been observed across various ethnic groups.

Purpose of the Study:

  • To investigate the association between pemphigus subtypes and HLA genes in the Moroccan population for the first time.
  • To identify specific HLA alleles and haplotypes linked to pemphigus susceptibility or protection in Morocco.

Main Methods:

  • Compared 52 unrelated Moroccan pemphigus patients with 178 healthy controls.
  • Utilized complement-dependent microlymphocytotoxic method for HLA class I typing.
  • Employed sequence-specific primer amplification for HLA class II typing.

Main Results:

  • No significant association found with HLA-A or HLA-B antigens.
  • Increased frequencies of DRB1*04, DRB1*14, and DQB1*03 alleles observed.
  • Decreased frequencies of DRB1*15 and DQB1*06 alleles noted, suggesting a protective effect for the DRB1*15-DQB1*06 haplotype.
  • DRB1*04-DQB1*03 and DRB1*14-DQB1*05 haplotypes linked to increased susceptibility.

Conclusions:

  • Results confirm a genetic predisposition to pemphigus in the Moroccan population.
  • Identified specific HLA alleles and haplotypes influencing pemphigus risk.
  • The observed susceptibility alleles are similar to those found globally, suggesting genetic factors alone do not fully explain pemphigus prevalence in Morocco.
