Researchers identified serine/threonine kinase 31 (STK31) as a novel cancer/testis antigen. STK31 is highly expressed in colorectal, gastric, and esophageal cancers, showing potential for gastrointestinal cancer immunotherapy.

Area of Science:

  • Oncology
  • Molecular Biology
  • Immunology

Background:

  • Advanced molecular biology tools aid cancer diagnosis and treatment.
  • Cancer/testis (CT) antigens are targets for cancer immunotherapy.
  • Identifying novel CT antigens is crucial for developing new cancer therapies.

Purpose of the Study:

  • To develop a new methodology for identifying novel cancer/testis (CT) antigens.
  • To identify and characterize a novel CT antigen expressed in colorectal cancer.
  • To evaluate the potential of the identified antigen as a target for cancer immunotherapy.

Main Methods:

  • Combined laser microdissection and cDNA microarray to analyze gene expression profiles.
  • Compared gene expression in colorectal cancer cells with normal colon epithelium and normal testis.

  • Utilized reverse transcription-polymerase chain reaction (RT-PCR) to validate gene expression levels.
  • Main Results:

    • Identified 38 candidate CT antigens using the novel methodology.
    • Discovered serine/threonine kinase 31 (STK31) as a novel CT antigen.
    • STK31 showed significantly higher expression in colorectal cancer tissues compared to normal tissues (P < 0.0001).
    • STK31 was also frequently expressed in gastric and esophageal cancers.
    • STK31 peptide successfully elicited specific cytotoxic T lymphocytes (CTLs) that lysed target cells.

    Conclusions:

    • The developed methodology effectively facilitates the identification of novel CT antigens.
    • STK31 is a promising candidate for cancer immunotherapy, particularly against gastrointestinal cancers.
    • STK31-specific CTLs demonstrate potential for targeted cancer treatment.
