Amyloid fibrils are aggregates of misfolded proteins.  Under most circumstances, misfolded proteins are either refolded by chaperone proteins or degraded by the proteasome. Summary

Cellular prion protein (PrP(C)) was detected in sheep urine. Elevated levels of this prion protein were found in scrapie-infected sheep, suggesting potential as a biomarker for transmissible spongiform encephalopathies.

Background:

  • Cellular prion protein (PrP(C)) misfolding causes transmissible spongiform encephalopathies.
  • PrP(C) is a biomarker for these neurodegenerative diseases.
  • PrP(C) is expressed in the central nervous system and found in peripheral tissues and bodily fluids like urine.

Purpose of the Study:

  • To characterize prion protein in urine samples from normal and scrapie-infected sheep.
  • To investigate the potential of urinary PrP(C) as a biomarker for scrapie.

Main Methods:

  • Urine samples from scrapie-infected and healthy sheep were collected.
  • Samples were concentrated and analyzed using Western blot with anti-PrP monoclonal antibodies.
  • Quantification of PrP(C) was performed by comparison with recombinant ovine PrP standards.

Main Results:

  • A protease K-sensitive PrP(C) band (27-30 kDa) was visualized in sheep urine.
  • Background PrP(C) levels in healthy sheep urine ranged from 0-0.16 ng/ml.
  • Seven of 33 scrapie-infected sheep showed elevated PrP(C) levels (0.3-4.7 ng/ml) in their urine.

Conclusions:

  • Prion protein is present in the urine of sheep.
  • Elevated urinary PrP(C) levels were observed in naturally scrapie-infected sheep.
  • Further research is needed to explore the origin and significance of urinary PrP(C) in scrapie infection.
