Background:

Bortezomib is a novel proteasome inhibitor.
It has demonstrated improved outcomes in multiple myeloma treatment.
Bortezomib exhibits manageable toxicities.

Purpose of the Study:

To review the chemistry, pharmacokinetics, and metabolism of bortezomib.
To evaluate the clinical efficacy and toxicity of bortezomib.
To assess bortezomib use in specific patient populations and its effects on bone metabolism.

Main Methods:

Literature search of bortezomib studies published within the last 10 years.
Inclusion of recent congress abstracts on bortezomib.

Main Results:

Bortezomib has improved response rates, overall survival, and time to progression in relapsed or refractory multiple myeloma.
It is effective as a single agent and in combination regimens.
Promising results are reported for bortezomib combinations in frontline induction therapy.

Conclusions:

Bortezomib expands therapeutic options for multiple myeloma.
It benefits patient subgroups for whom conventional therapies are unsuitable.
Further research supports bortezomib's role in managing this disease.
