Insights
Boronate affinity chromatography is superior for estimating hemoglobin A1c (HbA1c) in diabetic patients with hemoglobin D (HbD). This method minimizes interference, ensuring accurate glycemic control assessment in both heterozygous and homozygous HbD cases.
Area of Science:
- Clinical Chemistry
- Diabetology
- Chromatography
Background:
- Accurate hemoglobin A1c (HbA1c) measurement is crucial for managing diabetes.
- Hemoglobin variants, such as hemoglobin D (HbD), can interfere with standard HbA1c estimation methods.
- Type I and Type II diabetes patients with HbD require specialized diagnostic approaches.
Purpose of the Study:
- To compare the efficacy of ion exchange chromatography and boronate affinity chromatography for HbA1c estimation.
- To evaluate these methods in diabetic patients with hemoglobin D (both homozygous and heterozygous).
Main Methods:
- Evaluation of systems based on ion exchange chromatography.
- Evaluation of systems based on boronate affinity chromatography.
- Comparative performance analysis in patients with homozygous and heterozygous HbD disease.
Main Results:
- Boronate affinity chromatography demonstrated minimal interference from HbD.
- This interference was reduced in both heterozygous and homozygous HbD diabetic patients.
- Ion exchange chromatography showed greater interference from HbD.
Conclusions:
- Boronate affinity chromatography is a reliable method for HbA1c estimation in diabetic patients with HbD.
- This technique aids in the accurate evaluation of glycemic control in this patient population.
- It offers an advantage over ion exchange chromatography for HbA1c measurement in the presence of HbD.
Objectives:
To compare ion exchange and boronate affinity chromatography for HbA(1c) estimation in patients with type I and II diabetes having hemoglobin D.
Design And Methods:
Systems based on ion exchange and boronate affinity chromatography were evaluated and compared for their performance for HbA(1c) estimation in patients with homozygous and heterozygous D disease.
Results:
Boronate affinity chromatography shows least interference by HbD in heterozygous as well as homozygous diabetic patients for HbA(1c) estimation.
Conclusions:
The use of boronate affinity chromatography was found to be helpful in evaluating glycemic control in diabetic subjects with HbD.
