Flow Cytometry01:23

Flow Cytometry

The development of flow cytometry techniques began in 1934 with initial attempts by Andrew Moldavan, a bacteriologist who counted the cells in a flowing capillary system. Moldavan pumped cells through a capillary tube focused under a microscope for visualization. The invention of photometry allowed the measurement of differentially-stained cells, and Louis Kamentsky developed the first multiparameter flow cytometer in 1965 to identify and count the cancer cells in cervical tissue specimens.

Area of Science:

  • Biotechnology
  • Analytical Chemistry
  • Cell Biology

Background:

  • Flow cytometry is powerful for cell screening but inefficient for large-scale, multi-sample operations.
  • Modern drug discovery requires high-throughput screening of cellular targets against vast compound libraries.

Purpose of the Study:

  • To develop an automated sample handling method for high-throughput flow cytometry.
  • To enable efficient large-scale screening of discrete cell suspensions.

Main Methods:

  • Developed a flow injection analysis (FIA) approach for automated sample handling.
  • Implemented a plug flow system where discrete sample suspensions are injected into a carrier fluid.
  • Samples are sequentially delivered to the laser interrogation point of the flow cytometer.

Main Results:

  • The plug flow system successfully automates sample delivery for flow cytometry.
  • Precisely defined volumes of individual sample suspensions are sequentially processed.
  • Demonstrated the system's capability for high-throughput analysis.

Conclusions:

  • The developed flow injection analysis plug flow system enhances the efficiency of flow cytometry for large-scale screening.
  • This automated approach is beneficial for applications like drug discovery and broader biological investigations.
  • The system provides a foundation for advancing high-throughput cellular analysis.
