Immunogold Electron Microscopy01:20

Immunogold Electron Microscopy

Immunoelectron microscopy utilizes immunogold labeling of endogenous proteins with specific antibodies to detect and localize these proteins in cells and tissues. The procedure provides insights into the distribution and quantification of protein under different stimulation conditions offering clues about their functions. Conjugating highly electron-dense gold particles with primary or secondary antibodies allow antigen detection on and within cells, with high resolution and specificity.

Summary

A novel antibody targeting Zonula Occludens-2 (ZO-2) non-specifically labels Mauthner (M-) cells in fish. This discovery aids in identifying M-cells and their intricate processes for anatomical studies.

Area of Science:

  • Neuroscience
  • Cell Biology
  • Ichthyology

Background:

  • Mauthner (M-) cells are crucial for fish escape responses.
  • Investigating protein co-localization at mixed synapses is important for understanding neuronal function.

Purpose of the Study:

  • To explore the co-localization of scaffold and gap junction proteins at mixed synapses.
  • To identify the Mauthner cell and its fine processes using a specific antibody.

Main Methods:

  • Immunohistochemistry using an antibody against Zonula Occludens-2 (ZO-2).
  • Microscopy to observe cellular labeling patterns.

Main Results:

  • A specific antibody against ZO-2 labeled Mauthner cells.
  • The staining was evenly distributed along M-cell dendrites and axons, including fine processes.
  • The observed labeling was determined to be non-specific.

Conclusions:

  • The ZO-2 antibody recognizes a soluble protein primarily expressed in Mauthner cells.
  • This antibody can facilitate Mauthner cell identification in anatomical and immunohistochemical studies.
  • It offers an alternative to traditional methods like intracellular tracer injection.
