Electrochemically deposited Prussian blue (PB) films show enhanced stability on glassy carbon electrodes when treated with tetrabutylammonium toluene-4-sulfonate (TTS). This improved PB film stability enables a robust multi-enzyme biosensor for sensitive sucrose detection.
Area of Science:
- Electrochemistry
- Biosensor Technology
- Materials Science
Background:
- Prussian blue (PB) films are crucial for electrochemical applications but often suffer from poor stability.
- Enhancing the stability of PB films on glassy carbon (GC) electrodes is essential for reliable biosensor development.
- Coupling PB films with oxidase enzyme systems offers potential for sensitive analyte detection.
Purpose of the Study:
- To investigate the stabilization of electrochemically deposited Prussian blue (PB) films on glassy carbon (GC) electrodes.
- To develop and demonstrate a multi-enzyme biosensor for sucrose detection utilizing stabilized PB films.
- To evaluate the performance and stability of the developed sucrose biosensor.
Main Methods:
- Electrochemical deposition of Prussian blue films on glassy carbon electrodes.
- Treatment of electrodes with tetrabutylammonium toluene-4-sulfonate (TTS) to enhance PB film stability.
- Immobilization of a tri-enzyme system (glucose oxidase, mutarotase, invertase) onto PB-modified GC electrodes using glutaraldehyde and bovine serum albumin.
- Electrochemical detection of hydrogen peroxide generated by the enzyme cascade using flow injection analysis.
Main Results:
- Tetrabutylammonium toluene-4-sulfonate (TTS) treatment significantly enhanced the operational and storage stability of PB films.
- A tri-enzyme biosensor demonstrated linear responses for sucrose (4-800 μM), glucose (2-800 μM), and H2O2 (1-800 μM).
- The biosensor achieved low detection limits (4.5 μM for sucrose) and a high sample throughput (approx. 60 samples/h).
Conclusions:
- Electrochemical activation with TTS is an effective strategy for stabilizing Prussian blue films on GC electrodes.
- The developed multi-enzyme biosensor exhibits excellent sensitivity, stability, and efficiency for sucrose determination.
- This work highlights the potential of stabilized PB films in constructing advanced electrochemical biosensing platforms.
