Common type 2 diabetes risk gene variants associate with gestational diabetes

Gestational diabetes mellitus (GDM) shares genetic risk factors with type 2 diabetes. Genetic variants associated with type 2 diabetes increase GDM risk, suggesting a common underlying pathology.

- Most type 2 diabetes risk alleles showed an increased odds ratio for GDM, with the exception of the WFS1 rs10010131 variant.
- A combined analysis of all 11 variants revealed a significant additive effect on GDM risk (OR 1.18 per risk allele, P = 3.2 x 10(-6)).
- The area under the ROC curve was 0.62 for genetic testing alone and improved to 0.73 when combined with age and BMI.
Conclusions:
- The prevalence of type 2 diabetes risk alleles in women with prior GDM supports their role in GDM susceptibility.
- These findings suggest that GDM and type 2 diabetes may share common underlying genetic factors, potentially representing different manifestations of the same disease entity.
