Background:

  • Ceratostigma willmottianum, commonly known as Radix Ceratostigmae, is a medicinal plant.
  • Plumbagin is a key active compound found in Radix Ceratostigmae.

Purpose of the Study:

  • To establish validated qualitative and quantitative analytical methods for Ceratostigma willmottianum.
  • To ensure the accurate identification and precise determination of plumbagin content.

Main Methods:

  • Thin-Layer Chromatography (TLC) was employed for the qualitative identification of Radix Ceratostigmae, using authentic material and plumbagin as a reference standard.
  • High-Performance Liquid Chromatography (HPLC) was utilized for the quantitative determination of plumbagin content in Radix Ceratostigmae extracts.

Main Results:

  • The TLC method demonstrated simplicity and specificity for identifying Radix Ceratostigmae.
  • The HPLC method, using a Diamonsil C18 column with a methanol-water mobile phase (75:25), effectively separated plumbagin.
  • The quantitative analysis showed a linear range for plumbagin from 33.6-313.6 ng, with an average recovery of 100.3% and a relative standard deviation (RSD) of 1.9%.

Conclusions:

  • The developed TLC and HPLC methods are suitable for the reliable identification and quantification of plumbagin in Ceratostigma willmottianum.
  • These validated methods contribute to the quality control and standardization of C. willmottianum in pharmaceutical applications.
