This study shows that Dissociation-enhanced lanthanide fluoro immunoassay (DELFIA) reliably detects lipopolysaccharide (LPS) by measuring interleukin-6 (IL-6) secretion. Further validation is recommended for this sensitive immunoassay method.
Area of Science:
- Immunology
- Biochemistry
- Cell Biology
Background:
- Lipopolysaccharide (LPS) is a critical component of Gram-negative bacteria, triggering potent immune responses.
- Interleukin-6 (IL-6) is a key pro-inflammatory cytokine involved in the host's response to LPS.
- Accurate measurement of LPS is essential for diagnosing infections and monitoring endotoxemia.
Purpose of the Study:
- To evaluate the efficacy of Dissociation-enhanced lanthanide fluoro immunoassay (DELFIA) for measuring LPS-induced IL-6 secretion.
- To assess the reliability and sensitivity of the DELFIA method in detecting LPS.
- To compare DELFIA with a bioassay for LPS detection.
Main Methods:
- Utilized Mono Mac 6 cells, a human monocytic cell line, for IL-6 secretion assays.
- Employed Dissociation-enhanced lanthanide fluoro immunoassay (DELFIA), a time-resolved fluorescence-based technique.
- Compared DELFIA results with those obtained from the B9 bioassay.
Main Results:
- DELFIA demonstrated a strong correlation with the B9 bioassay for LPS detection.
- The DELFIA assay exhibited no cross-reactivity with various human cytokines or murine IL-6.
- The detection limit for LPS was 2.5 pg/ml in aqueous solution and 25 pg/ml in a 25% total parenteral nutrition (TPN) product.
- TPN concentrations above 25% inhibited IL-6 secretion from Mono Mac 6 cells, though it did not interfere with the DELFIA assay itself.
Conclusions:
- The combination of Mono Mac 6 cells and DELFIA provides a reliable method for LPS detection.
- Further validation of this DELFIA-based method by independent laboratories is warranted.
- This approach offers a sensitive and specific tool for assessing LPS presence and its biological effects.
