Background:

Purpose of the Study:

  • To establish the most effective protocol for purifying eosinophils from nasal polyp tissue.
  • To compare different purification methods for their efficiency, purity, and cell viability.
  • To enable future in vitro studies on tissue-resident eosinophils.

Main Methods:

  • Nasal polyps were minced and enzymatically digested to create a cell suspension.
  • Two purification techniques were evaluated: positive magnetic selection (CCR3+) and negative magnetic selection (CD3/CD14/CD16).
  • Purified eosinophils were cultured with cytokines (IL-3, IL-5, GM-CSF) and assessed for viability via flow cytometry.

Main Results:

  • Tissue-derived eosinophils showed comparable surface receptor expression to peripheral blood eosinophils.
  • Positive selection yielded high purity (>90%) but lower viability (63%).
  • Negative selection resulted in lower purity (66%) but significantly higher viability (88%), suitable for in vitro activation.

  • Eosinophils can be successfully purified from nasal polyp tissue.
  • Negative selection is advantageous for preserving eosinophil viability, facilitating in vitro culture and activation.
  • This method advances the study of tissue eosinophils in inflammatory conditions.
