Summary

L-histidine enhances zebrafish learning and stress response, while thioperamide impairs it. L-histidine also mitigates stress-induced blood glucose increases, suggesting a key role for the histaminergic system.

Area of Science:

  • Neuroscience
  • Behavioral Science
  • Pharmacology

Background:

  • The histaminergic system plays a role in cognitive functions.
  • Stress can negatively impact learning and memory.
  • Histamine H3 receptors are involved in neurotransmitter release and modulation.

Purpose of the Study:

  • To investigate the effects of L-histidine and thioperamide on zebrafish learning under stress.
  • To explore the role of the histaminergic system in learning and stress modulation.
  • To assess the impact of L-histidine on stress-induced physiological changes.

Main Methods:

  • Zebrafish were trained to associate a stimulus with a reward over five days.
  • Animals received intraperitoneal injections of L-histidine, thioperamide, or saline.
  • Confinement stress was applied to a subset of animals before learning tasks.
  • Learning was quantified by measuring the time to approach a food source.
  • Blood glucose levels were measured after stress exposure.

Main Results:

  • Saline-treated zebrafish demonstrated learning, indicated by decreased approach times.
  • Confinement stress delayed learning in saline-treated zebrafish.
  • L-histidine facilitated learning in both stressed and non-stressed zebrafish.
  • Thioperamide inhibited learning in both non-stressed and stressed zebrafish.
  • L-histidine administration prevented the rise in blood glucose levels post-stress.

Conclusions:

  • The histaminergic system, influenced by L-histidine and H3 receptor activity, significantly impacts learning in zebrafish.
  • L-histidine acts as a cognitive enhancer and stress modulator in zebrafish.
  • Thioperamide, an H3 receptor antagonist, impairs learning processes.
  • These findings highlight the therapeutic potential of targeting the histaminergic system for cognitive enhancement and stress management.
