Summary
Adipose tissue renin expression and secretion are regulated during cell differentiation. Forskolin and TNFalpha influence renin mRNA and protein release, with distinct mechanisms observed in 3T3-L1 adipocytes.
Area of Science:
- Biochemistry
- Cell Biology
- Endocrinology
Background:
- Adipose tissue has a local renin-angiotensin system, but renin synthesis and release are not well understood.
- Investigating renin regulation in adipocytes is crucial for understanding local tissue function.
Purpose of the Study:
- To investigate the expression and secretion of renin during 3T3-L1 adipocyte differentiation.
- To determine the effects of forskolin and TNFalpha on renin regulation in mature adipocytes.
Main Methods:
- Utilized differentiating 3T3-L1 cells to analyze renin mRNA and protein activity.
- Treated differentiated adipocytes with forskolin, TNFalpha, IL-6, insulin, and Angiotensin II.
- Assessed the impact of lipase and PKA inhibitors on renin secretion and mRNA levels.
- Compared renin expression in obese mice epididymal adipose tissue to lean controls.
Main Results:
- Renin was undetectable in preadipocytes but upregulated upon differentiation.
- Forskolin increased renin mRNA 28-fold and secretion 12-fold; TNFalpha decreased mRNA fourfold but increased secretion sevenfold.
- Lipase inhibitor (E600) and PKA inhibitor (H89) attenuated forskolin-induced renin release.
- Obese mice showed reduced adipose tissue renin expression, consistent with TNFalpha effects.
Conclusions:
- Renin expression and secretion in adipocytes are complex and influenced by hormonal and metabolic factors.
- Forskolin and TNFalpha differentially regulate renin mRNA and protein secretion via distinct pathways.
- Findings suggest a role for adipose tissue renin in metabolic regulation and obesity.

Adrenergic Receptors: β Subtype
β-adrenoceptors have varied sensitivities towards adrenaline, noradrenaline, and isoprenaline. The order of agonist potency is as follows:
Isoprenaline > Adrenaline > Noradrenaline
Neurotransmitter binding to these receptors causes activation of adenylyl cyclase resulting in increased concentrations of cAMP and modulation of calcium ion channels within the cell. They are further classified into β1, β2, and β3 subtypes.
β1-adrenoceptors: β1-adrenoceptors have equal affinities for...

Hormonal Regulation
The renin-aldosterone system is an endocrine system which guides the renal absorption of water and electrolytes, thus managing blood pressure and osmoregulation. Activation of the system begins in the kidneys with a small cluster of cells adjacent to the afferent and efferent blood vessels of the renal corpuscle. As the nephrons are filtering blood, juxtaglomerular cells monitor blood pressure. If they detect a decrease in pressure, they release the hormone renin into the bloodstream.
