Area of Science:

  • Biomedical imaging
  • Surgical oncology
  • Medical diagnostics

Background:

  • Methylene blue (MB) is a widely used, safe, and cost-effective dye for sentinel lymph node (SLN) mapping.
  • Current methods often require surgical intervention to visualize the blue-dyed SLN in deep tissues.

Purpose of the Study:

  • To investigate the fluorescence properties of methylene blue (MB).
  • To demonstrate MB's utility for near-infrared (NIR) fluorescence mapping of SLNs.

Main Methods:

  • Utilized a fluorescence spectrometer and a wavelength-resolved fluorescence spectral in vivo imaging system.
  • Administered MB intradermally into rabbit breast tissue.
  • Observed lymphatic flow and axillary SLNs via emitted NIR fluorescence.

Main Results:

  • MB demonstrated near-infrared (NIR) fluorescence properties suitable for SLN mapping.
  • Intradermal injection allowed direct visualization of lymphatic flow and SLNs through NIR fluorescence in deep tissue.
  • Fluorescence was retained in the SLN for several hours without radioactivity.

Conclusions:

  • Methylene blue (MB) can be effectively used for NIR fluorescence-guided sentinel lymph node (SLN) mapping.
  • This technique allows non-surgical, direct visualization of SLNs in deep tissues.
  • MB NIR fluorescence mapping offers significant advantages over traditional SLN identification methods.
