Abnormal Proliferation

Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. The Pituitary Gland

The pituitary is a small endocrine organ in the sphenoid bone under the hypothalamus. Primarily, the pituitary in adults has two distinct anatomical and functional regions— the anterior and posterior lobes. During human fetal development, a third pituitary gland region called the pars intermedia atrophies and disappears. However, some of its cells migrate and exist adjacent to the anterior pituitary in adults.

Background:

  • Rab3B is a GTP-binding protein involved in exocytosis regulation.
  • Its role in normal human pituitary and pituitary adenomas requires further elucidation.

Purpose of the Study:

  • To investigate the presence and distribution of Rab3B in various human pituitary adenomas.
  • To assess the impact of specific therapeutic agents on Rab3B expression in pituitary tumors.

Main Methods:

  • Immunohistochemical analysis of 130 surgically removed human pituitary adenomas.
  • Inclusion of 10 non-tumorous adenohypophyses for comparison.
  • Tumor classification based on WHO criteria (histologic, immunohistochemical, ultrastructural).

Main Results:

  • Rab3B immunopositivity was strongest in corticotroph adenomas, followed by thyrotroph, lactotroph, gonadotroph, null cell, and somatotroph adenomas.
  • Bromocriptine treatment correlated with decreased Rab3B immunoexpression; octreotide had no significant effect.
  • Cytoplasmic and evenly distributed Rab3B staining was observed, with no staining in normal adenohypophyses.

Conclusions:

  • Rab3B is differentially expressed across pituitary adenoma subtypes.
  • Rab3B is implicated in the regulation of pituitary hormone secretion.
  • Therapeutic interventions can modulate Rab3B expression in pituitary tumors.
