Summary
Peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1alpha) is essential for exercise to increase vascular endothelial growth factor (VEGF) in muscles. AMPK activation also increases VEGF via PGC-1alpha.
Area of Science:
- Exercise physiology
- Molecular biology
- Skeletal muscle biology
Background:
- Vascular endothelial growth factor (VEGF) plays a crucial role in angiogenesis and muscle adaptation to exercise.
- The role of PGC-1alpha and AMPK in exercise-induced VEGF expression, particularly in aging muscle, remains incompletely understood.
Purpose of the Study:
- To investigate the necessity of PGC-1alpha for exercise-induced VEGF expression in young and old mice.
- To determine if AMPK activation enhances VEGF expression through a PGC-1alpha-dependent pathway.
Main Methods:
- Utilized whole-body PGC-1alpha knockout (KO) and wild-type (WT) mice.
- Administered exercise training (5 weeks or lifelong), single exercise bouts, and AICAR injections.
- Measured VEGF protein and mRNA expression, and capillary-to-fiber ratio in skeletal muscle.
Main Results:
- PGC-1alpha KO mice exhibited significantly lower basal VEGF protein and capillary-to-fiber ratio compared to WT mice.
- Acute exercise and AICAR treatment increased VEGF mRNA in WT but not in KO mice.
- Exercise training increased VEGF protein in WT mice, an effect absent in KO mice, and prevented age-related decline in WT but not KO mice.
- Repeated AICAR treatment increased VEGF protein in WT but not KO mice.
Conclusions:
- PGC-1alpha is indispensable for exercise-induced upregulation of skeletal muscle VEGF expression.
- PGC-1alpha mediates the prevention of age-associated decline in VEGF protein content due to training.
- Findings suggest an AMPK-mediated regulation of VEGF expression that is dependent on PGC-1alpha.

Regulation of Angiogenesis and Blood Supply
Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits. Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl hydroxylase and factor...
TGF - β Signaling Pathway
The TGF-β signaling pathway regulates cell growth, differentiation, adhesion, motility, and development. TGF-β ligands that induce TGF-β signaling are synthesized in their latent form. Several proteases or cell surface receptors such as integrins act upon the latent form, releasing the active ligand. There are three types of mammalian TGF-βs: (TGF-β1, TGF-β2, and TGF-β3) that bind as homodimers or heterodimers to TGF-β receptors. The TGF-β receptors are of three kinds RI, RII, and RIII. The RI...
GPCRs Regulate Adenylyl Cylase Activity
Some GPCRs transmit signals through adenylyl cyclase (AC), a transmembrane enzyme. AC helps synthesize second messenger cyclic adenosine monophosphate (cAMP). AC catalyzes cyclization reaction and converts ATP to cAMP by releasing a pyrophosphate. The pyrophosphate is further hydrolyzed to phosphate by the enzyme pyrophosphatase, which drives cAMP synthesis to completion. However, cAMP is rapidly degraded to 5′ AMP by the enzymes phosphodiesterase (PDE), preventing overstimulation of cells.
