Ribosome Profiling02:24

Ribosome Profiling

Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

  • Primate Research

Background:

  • Gene microarray analysis offers high-throughput gene expression profiling in non-human primates.
  • Rhesus macaque gene annotation has evolved, impacting transcript identification precision.
  • Accurate gene expression data requires robust normalization and validation strategies.

Purpose of the Study:

  • To outline considerations for gene annotation and data interpretation using Affymetrix microarray platforms for rhesus macaques.
  • To highlight the importance of annotation revisions and their impact on transcript identification.
  • To discuss normalization methods and validation techniques for microarray data.

Main Methods:

  • Utilizing Affymetrix microarray platforms for gene expression profiling in rhesus macaques.
  • Analyzing gene annotation revisions and reassessments between March 2007 and November 2008.
  • Implementing normalization techniques for comparing gene expression across multiple chips.
  • Employing PCR-based methodology for external corroboration of microarray findings.

Main Results:

  • Annotation of the rhesus macaque genome array has undergone significant revisions, affecting probeset assignments.
  • Multiple annotation systems show substantial reassignment of probesets over time.
  • Appropriate normalization is crucial for accurate quantitation and comparison of gene expression levels.
  • Validation of internal reference genes is essential for normalizing PCR-based corroboration of microarray data.

Conclusions:

  • Effective gene expression profiling in rhesus macaques necessitates meticulous attention to gene annotation accuracy.
  • Proper normalization and validation are critical for reliable microarray data interpretation and external corroboration.
  • Availability of free tools aids researchers in normalization and reference gene selection for microarray studies.
