Purpose of the Study:

  • To investigate amygdala processing in individuals with untreated unipolar depression using functional MRI (fMRI).
  • To assess the relationship between amygdala activation, emotion recognition, and depression severity.

Main Methods:

  • Functional MRI (fMRI) was employed with a standardized emotion face recognition task.
  • Fourteen individuals with untreated major depression and 15 healthy controls were studied.
  • Voxel-level analysis and region of interest (ROI) analysis (bilateral amygdala) were performed, including correlation with HAM-D scores.

Main Results:

  • Both groups showed increased amygdala activity when viewing emotive faces versus geometric shapes.
  • Depressed individuals exhibited significantly greater amygdala activation, particularly on the left side.
  • Positive correlations were found between amygdala activation and Hamilton Depression Rating Scale (HAM-D) scores in patients.

Conclusions:

  • This study provides in vivo neuroimaging evidence of abnormal amygdala functioning in unipolar depression.
  • Heightened amygdala reactivity to emotional stimuli may be a neural correlate of major depressive disorder.
