Enzyme-Linked Immunosorbent Assay01:33

Enzyme-Linked Immunosorbent Assay

In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.
There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen.  Enzyme-substrate reaction allows the antigen to be visualized or quantified.
Automated Microbial Diagnostics01:24

Automated Microbial Diagnostics

Automated diagnostic analyzers have transformed clinical microbiology by providing rapid and reliable methods for pathogen identification and antibiotic susceptibility testing. Among these systems, the Vitek 2 is widely used because it automates the traditionally labor-intensive processes of microbial identification (ID) and antibiotic susceptibility testing (AST), delivering standardized and timely results that are essential for effective patient care.Microbial Identification with ID CardsThe...

Area of Science:

  • Biomedical Engineering
  • Analytical Chemistry
  • Point-of-Care Diagnostics

Background:

  • Microfluidic devices offer miniaturized platforms for complex biological assays.
  • Developing cost-effective and user-friendly diagnostic tools is crucial for widespread point-of-care (POC) applications.
  • Existing immunoassay formats can be complex and require specialized laboratory equipment.

Purpose of the Study:

  • To design and demonstrate an inexpensive, disposable, luminescence-based microfluidic immunoassay cassette.
  • To integrate this cassette into a functional point-of-care diagnostic system.
  • To evaluate the cassette's performance with different detection modalities and clinically relevant analytes.

Main Methods:

  • Fabrication of a two-part microfluidic cassette with integrated reagent storage and finger-actuated fluid control.
  • Sequential actuation of pouches to drive reagent and sample flow for immunoassay processes.
  • Integration of both immunochromatographic and microbead-based detection methods within the cassette.
  • Evaluation of cassette performance using HIV antibodies in saliva and the chemokine IL-8.

Main Results:

  • Demonstrated successful fluid motion driven by finger-actuated pouches.
  • Constructed and evaluated both immunochromatographic and microbead-based detection cassettes.
  • Achieved detection of HIV antibodies in saliva samples with reasonable sensitivity and good repeatability.
  • Successfully detected the proinflammatory chemokine IL-8 using the microbead-based cassette.

Conclusions:

  • The developed microfluidic immunoassay cassette is a viable platform for point-of-care diagnostics.
  • The design allows for storage of liquid reagents and user-friendly operation via finger-actuation.
  • The system demonstrates compatibility with multiple detection methods and shows promise for sensitive and repeatable analyte detection.
