Certain genetic variations in chemokine genes may influence the risk of non-Hodgkin lymphoma (NHL). The CCR5 Delta32 deletion, for example, appears to reduce NHL risk in men.

Area of Science:

  • Genetics
  • Immunology
  • Oncology

Background:

  • Polymorphisms in chemokine genes are linked to HIV-related lymphoma.
  • Their role in non-HIV-related lymphoma remains understudied.

Purpose of the Study:

  • To investigate associations between NHL and gene polymorphisms/haplotypes.
  • Focus on CCR5, CCR2, CCL5, CXCL12, and CX(3)CR(1) in HIV-negative individuals.

Main Methods:

  • Pooled analysis of three case-control studies (N=1610 cases, N=1992 controls).
  • Used adjusted unconditional logistic regression for risk estimation.
  • Analyzed HIV-negative, non-Hispanic Caucasian populations.

Main Results:

  • The CCR5 Delta32 deletion reduced NHL risk in men (OR=0.56), particularly for diffuse large-cell and follicular lymphoma (FL).

  • A CCR2/CCR5 haplotype with CCR5 Delta32 also reduced NHL risk in men.
  • The CCL5 -403A allele was associated with reduced risk of FL and chronic lymphocytic leukemia/small lymphocytic lymphoma.
  • Conclusions:

    • Specific chemokine gene polymorphisms may modify NHL risk in HIV-negative individuals.
    • Findings suggest a potential protective role for CCR5 Delta32 and CCL5 -403A in certain NHL subtypes, particularly in men.
    • Further research is needed to validate these associations.
