[Changes in skin enzyme activity in experimental burns].

    Summary

Enzyme activities, including cathepsins and lactate dehydrogenase, decreased significantly after thermal burns in rats. Restoration of proteolytic enzyme activity in surrounding tissues may worsen burn wound depth.

    Area of Science:

    • Biochemistry
    • Wound Healing Research
    • Dermatology

    Background:

    • Thermal burns cause significant tissue damage and inflammation.
    • Enzyme activity changes are critical indicators of tissue injury and repair processes.

    Purpose of the Study:

    • To investigate the dynamic changes in proteolytic enzyme and lactate dehydrogenase activities following thermal burns.
    • To correlate enzyme activity alterations with burn wound progression and healing stages.

    Main Methods:

    • Assay of cathepsins B and D, trypsin-like proteases, and leucine aminopeptidase activities.
    • Measurement of lactate dehydrogenase and its isoenzymes in burn-affected tissues.
    • Comparative analysis of enzyme levels in burn eschar, surrounding zones, and intact skin.

    Main Results:

    • A marked decrease in studied enzyme activities was observed within 24 hours post-burn.
    • Enzyme activities remained low for three weeks, followed by a gradual restoration.
    • Elevated proteolytic enzyme activity in tissues under the crust and in the demarcational zone correlated with burn wound deepening.

    Conclusions:

    • Initial decrease in enzyme activity suggests acute tissue injury and metabolic suppression.
    • Restored and elevated proteolytic enzyme activity in specific zones may contribute to delayed healing and deeper wound formation.
    • Understanding these enzymatic shifts is crucial for developing targeted burn treatment strategies.
