Objective:
To optimize the extraction technology and to establish the determination method of polysaccharides from Caulis Marsdeniae Tenacissimae.
Methods:
Phenol-sulphate colorimetry was used to determine the content of polysaccharides. According to the yield of polysaccharides, the orthogonal design was used to select the extraction technology of polysaccharides.
Results:
The optimum extraction procedure was 8 folds of water, refluxing and extracting for 3 times at 100 degrees C, and 1 h for each time. The concentration of polysaccharide was linear in the range of 0.004-0.014 mg/mL The average recovery was 99.72%, RSD = 0. 63%.
Conclusion:
The optimum extraction procedure is simple and reliable which is suitable for industrial production. The method for determination is simple, specific and accurate which can provide a method for mass control of Marsdenia tenacissima.
