Summary

Bovine Toll/interleukin-1 receptor domain-containing protein 8 (TIR8) is expressed in the gastrointestinal tract and kidney, similar to other mammals. This study identified TIR8 in additional bovine tissues and revealed multiple isoforms, suggesting complex regulation.

Area of Science:

  • Immunology
  • Molecular Biology
  • Veterinary Science

Background:

  • Toll/interleukin-1 receptor domain-containing protein 8 (TIR8), also known as SIGIRR, is an orphan receptor.
  • TIR8 is thought to negatively regulate Toll-like receptor (TLR) and IL-1 receptor (IL-1R) signaling, playing a role in inflammation modulation.
  • The expression pattern of TIR8 in bovine tissues was previously unknown, hindering understanding of its role in cattle.

Purpose of the Study:

  • To investigate the tissue distribution of TIR8 expression in non-pathologic bovine tissues.
  • To identify potential tissue-specific isoforms of bovine TIR8.
  • To provide insights into the function of TIR8 in cattle, particularly concerning gastrointestinal diseases.

Main Methods:

  • Northern blot analysis to detect TIR8 mRNA.
  • Qualitative and quantitative Real-Time PCR to confirm and quantify TIR8 mRNA expression.
  • Western blot immunodetection using a validated anti-human TIR8 polyclonal antibody to identify protein presence and isoforms.

Main Results:

  • Bovine TIR8 mRNA and protein were detected in the gastrointestinal tract and kidney, consistent with findings in humans and mice.
  • TIR8 expression was also identified in bovine lymph nodes, thymus, and thyroid gland.
  • Multiple TIR8 isoforms were observed in several bovine organs, indicating potential tissue-specific post-translational modifications.

Conclusions:

  • Bovine TIR8 is expressed in key immune and inflammatory-related tissues, including the GI tract, kidney, lymph nodes, thymus, and thyroid.
  • The presence of multiple TIR8 isoforms suggests complex regulatory mechanisms and diverse functional roles in cattle.
  • Further research is warranted to elucidate the specific functions of TIR8 and its isoforms in bovine health and disease.
