Chromatin Immunoprecipitation- ChIP

Chromatin immunoprecipitation, or ChIP, is an antibody-based technique used to identify sites on DNA that bind to transcription factors of interest or histone proteins. It also helps determine the type of histone modifications such as acetylation, phosphorylation, or methylation.

Background:

  • Seseli praecox is an endemic plant species from Sardinia belonging to the Apiaceae family.
  • Investigating endemic flora can lead to the discovery of novel bioactive compounds.

Purpose of the Study:

  • To isolate and characterize chemical constituents from the stems of Seseli praecox.
  • To evaluate the in vitro antiproliferative activity of a newly identified compound against cancer cell lines.

Main Methods:

  • Chemical investigation using chromatographic and spectroscopic techniques.
  • In vitro screening of the isolated compound against four human cancer cell lines.

Main Results:

  • An isopropenylated chromone (5-hydroxy-6-(2-Z-butenyl-3-hydroxymethyl)-7-methoxy-2-methylchromone) was isolated and identified.
