Summary

Diazepam did not acquire anxiogenic properties in rats, as conditioning with a tone signal did not induce anxiety. A compensatory anxiogenic response to diazepam was also not elicited in a second experiment.

Area of Science:

  • Neuroscience
  • Behavioral Pharmacology

Background:

  • Anxiogenic properties of drugs can be acquired through classical conditioning.
  • Diazepam is a commonly used anxiolytic medication.

Purpose of the Study:

  • To investigate if diazepam can acquire anxiogenic properties through association with an aversive event.
  • To determine if a small dose of diazepam can elicit a compensatory anxiogenic response.

Main Methods:

  • Rats were trained in the pentylenetetrazol (PTZ) model of anxiety in an operant chamber.
  • Experiment 1 involved classical conditioning with diazepam alone or diazepam plus a tone signal.
  • Experiment 2 tested for a compensatory anxiogenic response to a small dose of diazepam.

Main Results:

Related Experiment Videos

  • Rats conditioned with diazepam and a tone selected the anxiogenic-associated lever more often, indicating the tone induced anxiety, not diazepam.
  • No differences in lever selection were observed in the second experiment, suggesting no compensatory anxiogenic response to diazepam.

Conclusions:

  • Diazepam does not appear to acquire anxiogenic properties through signaling an aversive event.
  • A compensatory anxiogenic conditional response to diazepam was not elicited in this study.
