Assessment of nonhuman primate vaccine studies using simian immunodeficiency virus (SIV) models.

Main Results:

The Thai HIV vaccine trial showed diminishing efficacy over time, with greatest effect in low-risk individuals.
Two novel monoclonal antibodies demonstrated enhanced potency and broader reactivity.
A DNA prime/modified vaccinia virus Ankara boost regimen reduced SIV acquisition in macaques.
Attenuated SIV vaccines induced cellular immune responses controlling viral replication.
Cytomegalovirus-expressing SIV antigens provided robust protection against SIVmac239 challenge.

Conclusions:

Advancements in neutralizing antibodies and SIV vaccine models offer renewed optimism for HIV vaccine development.
Further research is needed to understand waning vaccine immunity and optimize delivery strategies.
Novel approaches, including SIV antigen-expressing viral vectors, show significant protective potential.
