1Laboratory for Advanced Research in Natural and Synthetic Chemistry, V. G. Vaze College, Mumbai University, Mithagar road, Mulund (East), Mumbai 400 081, India.
Andrographolide derivatives were synthesized and tested for anticancer properties. Compounds 3-5 showed significant activity against non-small cell lung cancer cells, indicating potential for new cancer therapies.
Area of Science:
- Medicinal Chemistry
- Pharmacology
- Oncology
Background:
- Andrographolide 1, a natural compound from Andrographis paniculata, exhibits antitumor effects.
- Existing research shows andrographolide's efficacy in breast cancer and leukemia models.
- There is a need for novel therapeutic agents against non-small cell lung cancer (NSCLC).
Purpose of the Study:
- To semi-synthesize novel andrographolide derivatives.
- To evaluate the in vitro cytotoxic activity of these derivatives against the A549 NSCLC cell line.
Main Methods:
- Semi-synthesis of andrographolide derivatives.
- In vitro cytotoxicity assays using the A549 (ATCC) cell line.
- Determination of half-maximal inhibitory concentration (IC50) values.
Main Results:
- Several andrographolide derivatives were successfully synthesized.
- Compounds 3-5 demonstrated potent in vitro activity against A549 cells.
- IC50 values for compounds 3-5 ranged from 22-31 microg/mL.
Conclusions:
- The synthesized andrographolide derivatives possess significant cytotoxic activity against NSCLC cells.
- Compounds 3-5 are promising candidates for further investigation as potential anti-NSCLC agents.
- This study highlights the therapeutic potential of modified andrographolide structures in cancer treatment.
