Interleukin 10 gene polymorphism in Iranian patients with multiple sclerosis

Interleukin-10 (IL-10) gene polymorphisms are not associated with multiple sclerosis (MS) in the Iranian population. This finding suggests IL-10 variants do not influence MS susceptibility or disease progression in this ethnic group.

Background:
- Interleukin-10 (IL-10) is a cytokine that modulates immune responses, including the inhibition of T-helper 1 (Th1) and T-helper 2 (Th2) cells.
- Genetic variations in the IL-10 promoter region may influence its expression levels and, consequently, immune system regulation.
Purpose of the Study:
- To investigate the association between specific single-nucleotide polymorphisms (SNPs) in the IL-10 promoter gene and the risk of developing multiple sclerosis (MS).
- To determine if IL-10 gene polymorphisms influence disease subtypes or progression in an Iranian MS patient cohort.
Main Methods:
- Genotyping of three IL-10 promoter SNPs (-1082, -819, -592) using Amplification Refractory Mutation System (ARMS) and PCR-Restriction Fragment Length Polymorphism (PCR-RFLP).
- Analysis of genetic data from 110 Iranian MS patients and 100 age- and ethnically-matched healthy controls.
Main Results:
- No statistically significant differences were observed in the allelic or genotype frequencies of the studied IL-10 polymorphisms between MS patients and controls.
- IL-10 gene polymorphisms did not show any association with MS susceptibility in the investigated Iranian population.
- Disease course, gender, and progression index did not correlate with the distribution of IL-10 alleles or genotypes.
Conclusions:
- IL-10 promoter polymorphisms are not significantly associated with multiple sclerosis in the Iranian population.
- These genetic variations do not appear to influence MS susceptibility or disease progression in this non-European cohort.
- Further research in diverse ethnic groups may be warranted to fully elucidate the role of IL-10 genetics in MS.
