Summary

Isobaric tags for relative and absolute quantification (iTRAQ) enable precise protein quantification in proteomics. This method labels peptides, allowing for sensitive and accurate relative protein abundance measurements through reporter ion analysis.

Area of Science:

  • Proteomics
  • Analytical Chemistry
  • Biochemistry

Background:

  • Protein relative quantification is crucial for proteomics.
  • Various methods exist, including isobaric tagging.

Purpose of the Study:

  • To describe the use of isobaric tags for relative and absolute quantification (iTRAQ).
  • To detail the procedure for labelling and analyzing peptide/protein samples using iTRAQ.

Main Methods:

  • Chemical tags (iTRAQ) attach to peptides via free amines.
  • Labeled samples are pooled and analyzed simultaneously using mass spectrometry.
  • Reporter ion intensity ratios indicate relative peptide proportions.

Main Results:

  • iTRAQ offers moderate sensitivity increase by summing signals from isobaric peptides.
